Do you usually have to put a deposit on a rental car?

I'll be either 21 or 22 (not sure of the exact date of my trip, it's right around my birthday) and it'll be the first time I rent a car. I was thinking booking it on orbitz but I'm not sure yet. Somebody mentioned to me I might have to put a couple hundred dollars cash deposit down? Is this true even if I pay with a debit or credit card?

A deposit is almost always the case. Use your credit card. Do not use your debit card. If you use your debit card, a deposit can tie up your available cash for withdrawl for several days even after you turn the car in. Plus, the credit card might provide you with some collision protection not offered under your debit card. But check in advance on the minimum age. Some rental agencies only rent to age 25 or older.
